If the ensemble relied on me to give the tuning pitch, I would use a tuner to provide it. If not, I would use my ear. In most orchestras, the oboe has that job. I have found that many orchestras play a little on the sharp side, so trying to line your tuba up with your tuner can sometimes get you into trouble. Sometimes, I prefer to use a tuner to get my slides set properly and then adjust the fundamental by ear.
