If your horn doesn't have any leaks and it's just one particular note that's way out of whack, try the following:
Borrow a sousaphone bit or two and insert one after your mouthpiece. Re-tune and see if that improves things any. If it does, try 2 tuning bits. If not, you're probably up a creek without major surgery.
The tuning bit trick often goes a long way toward fixing the flat third partial on Conn 2XJ horns.
